The modern lifestyle of humans is leading to a limited exposure to nature. While several wild species are adapting and thriving in anthropic environments, natural history knowledge is declining, and positive attitudes and behaviours towards nature are facing challenges. Because anticipating attitudes and engendering broad-based support for nature-related measures requires a good grasp of social contexts, we set out to evaluate the sociodemographic factors driving the perception, attitudes towards, and natural history knowledge of a keystone species-the European hedgehog. In 2022, we conducted a questionnaire answered by 324 Portuguese adults. We found generally positive feelings and attitudes towards this species. A higher degree of academic qualifications and previous personal experience with the species seem to play a role in (i) people's perception about human impacts on hedgehogs and (ii) positive attitudes, especially during encounters where the animals were in difficulty. Despite this, the extent of natural history knowledge was low overall, and the study population was self-aware of this. Our insights underline the need to tailor educational programmes if we are to encourage people to re-establish meaningful connections with nature, to foster social support for biodiversity stewardship, and to implement the One Health approach in a way that resonates with distinct social groups.

In recent years, implicit neural representations (INR) have shown their great potential to solve many computer graphics and computer vision problems. With this technique, signals such as 2D images or 3D shapes can be fit by training multi-layer perceptrons (MLP) on continuous functions, providing many advantages over conventional discrete representations. Despite being considered a promising approach to 2D image encoding and compression, the application of INR to image collections remains a challenge, since the number of parameters needed rapidly grow with the number of images. In this paper, we propose a fully implicit approach to INR which drastically reduces the size of MLP models in multiple image representation tasks. We introduce the concept of implicit coordinate encoder (ICE) and show it can be used to scale INR with the image number; specifically, by learning a common feature space between images. Furthermore, we show that our method is valid not only for image collections but also for large (gigapixel) images by applying a "divide-and-conquer" strategy. We propose an auto-encoder deep neural network architecture, with a single ICE (encoder) and multiple MLP (decoders), which are jointly trained following a multi-task learning strategy. We demonstrate the benefits coming from ICE when it is implemented as a one-dimensional convolutional encoder, including a better performance of the downstream MLP models with an order of magnitude fewer parameters. Our method is the first one to make use of convolutional blocks in INR networks, unlike the conventional approach of using MLP architectures only. We show the benefits of ICE in two experimental scenarios: a collection of twenty-four small ( 768×512 ) images (Kodak dataset), and a single large ( 3072×3072 ) image (dwarf planet Pluto), achieving better quality than previous fully-implicit methods, using up to 50% fewer parameters.

INTRODUCTION: Postoperative nausea and vomiting is still a common complication. Serotonin receptor antagonists are commonly used in clinical practice for antiemetic prophylaxis. Interindividual variations in drug response, including single nucleotide polymorphisms, are related to pharmacokinetic and pharmacodynamic changes in these drugs and may lead to a poor therapeutic response. This study aimed to evaluate the influence of CYP2D6 isoenzyme and ABCB1 gene polymorphisms on the frequency of postoperative nausea and vomiting with the use of ondansetron or palonosetron. METHODS: A randomized, double-blind clinical trial including 82 women aged 60 years or over undergoing laparoscopic cholecystectomy was conducted. Patients were randomized to receive either ondansetron or palonosetron for postoperative nausea and vomiting prophylaxis. DNA was extracted from saliva. Genetic polymorphisms were analyzed by real-time polymerase chain reaction. The following polymorphisms were analyzed: rs3892097 C/T, rs1128503 A/G, rs16947 A/G, rs1065852 A/G, rs1045642 A/G, rs2032582 C/A, and rs20325821 C/A. RESULTS: Overall, vomiting, and severe nausea occurred in 22.5% and 57.5% of patients, respectively. In the palonosetron group, patients with the GG genotype (rs16947 A/G) experienced more severe nausea (p = 0.043). In the ondansetron group, patients with the AA genotype (rs16947 A/G) presented mild nausea (p = 0.034), and those with the AA genotype (rs1065852 A/G) experienced more vomiting (p = 0.034). CONCLUSION: A low antiemetic response was observed with ondansetron in the presence of the AA genotype (rs16947 A/G) and the AA genotype (rs1065852 A/G), and a low therapeutic response was found with palonosetron in the presence of the GG genotype (rs16947 A/G) in laparoscopic cholecystectomy. Thiamine deficiency is associated with cerebellar dysfunction; however, the consequences of thiamine deficiency on the electrophysiological properties of cerebellar Purkinje cells are poorly understood. Here, we evaluated these parameters in brain slices containing cerebellar vermis. Adult mice were maintained for 12-13 days on a thiamine-free diet coupled with daily injections of pyrithiamine, an inhibitor of thiamine phosphorylation. Morphological analysis revealed a 20% reduction in Purkinje cell and nuclear volume in thiamine-deficient animals compared to feeding-matched controls, with no reduction in cell count. Under whole-cell current clamp, thiamine-deficient Purkinje cells required significantly less current injection to fire an action potential. This reduction in rheobase was not due to a change in voltage threshold. Rather, thiamine-deficient neurons presented significantly higher input resistance specifically in the voltage range just below threshold, which increases their sensitivity to current at these critical membrane potentials. In addition, thiamine deficiency caused a significant decrease in the amplitude of the action potential afterhyperpolarization, broadened the action potential, and decreased the current threshold for depolarization block. When thiamine-deficient animals were allowed to recover for 1 week on a normal diet, rheobase, threshold, action potential half-width, and depolarization block threshold were no longer different from controls. We conclude that thiamine deficiency causes significant but reversible changes to the electrophysiology properties of Purkinje cells prior to pathological morphological alterations or cell loss. Thus, the data obtained in the present study indicate that increased excitability of Purkinje cells may represent a leading indicator of cerebellar dysfunction caused by lack of thiamine.

OBJECTIVE: The present study was designed to test the hypothesis that there is a reduction in the activity of the enzyme cytochrome c oxidase (Cox) in Alzheimer's disease (AD). METHODS: Systematic review of literature and meta-analysis were used with data obtained from the PubMed, Scopus, MEDLINE, Lilacs, Eric and Cochrane. The keywords were Alzheimer's AND Cox AND mitochondria; Alzheimer's AND Cox AND mitochondria; Alzheimer's AND complex IV AND mitochondria. A total of 1372 articles were found, 23 of them fitting the inclusion criteria. The data were assembled in an Excel spreadsheet and analysed using the RevMan software. A random effects model was adopted to the estimative of the effect. RESULTS: The data shows a significant decrease in the activity of the Cox AD patients and animal models. CONCLUSION: Cox enzyme may be an important molecular component involved in the mechanisms underlying AD. Therefore, this enzyme may represent a possible new biomarker for the disease as a complementary diagnosis and a new treatment target for AD.

Abstract Introduction: Obesity, characterized by the accumulation of excess body fat, can alter respiratory mechanics and compromise functional capacity. Given its increasing prevalence and the significant morbidity associated with the condition, it is important to investigate techniques that enable rapid, easy measurement of lung function and the possible correlation between obesity and functional capacity. Objective: To assess lung function and functional capacity in obese adults and determine whether there is a correlation between reduced peak expiratory flow and impaired functional capacity. Methods: 30 participants, distributed into two groups: obese (36 ± 13 years) and normal weight (31 ± 9 years) were evaluated based on anthropometric measurements, the six-minute walk test (6MWT) and peak expiratory flow (PEF). Results: The obese individuals showed reduced PEF (382 ± 99 vs. 497 ± 104 L/min, p < 0.01), walked shorter distances in the 6MWT (453 ± 37 vs. 617 ± 50 m, p < 0.01), had higher blood pressure and perceived exertion (p < 0.05) when compared to normal weight participants. Additionally, there was a positive significant association between expiratory flow and distance walked in the 6MWT (r = 0.635 and p < 0.001). Conclusion: Obese individuals exhibited lower PEF and walked shorter distances in the 6MWT, indicating a positive correlation between these two variables. In light of this outcome, the assessment methods used could contribute to improving analysis of respiratory and functional status in this population and aid in exercise prescription.


Resumo Introdução: A obesidade, caracterizada por acúmulo excessivo de tecido adiposo no organismo, pode alterar a mecânica ventilatória, trazendo prejuízos funcionais. Considerando a prevalência crescente da obesidade, bem como a grande morbidade associada a esta condição, torna-se relevante investigar técnicas de mensuração rápidas e fáceis da função ventilatória e a possível correlação com a capacidade funcional. Objetivo: Avaliar a função ventilatória e a capacidade funcional em adultos obesos e determinar se pico de fluxo expiratório reduzido está correlacionado com prejuízo da capacidade funcional. Métodos: 30 participantes, distribuídos em dois grupos: obeso (36 ± 13 anos) e eutrófico (31 ± 9 anos) foram avaliados por meio de medidas antropométricas, do teste de caminhada de seis minutos (TC6´) e da avaliação do pico de fluxo expiratório (PFE). Resultados: Os indivíduos obesos apresentaram reduzido PFE (382 ± 99 vs. 497 ± 104 L/min, p < 0,01) e percorreram menor distância no TC6´ (453 ± 37 vs. 617 ± 50 m, p < 0,01) com maior pressão arterial e percepção de esforço (p < 0,05) quando comparados aos eutróficos. Além disso, foi encontrada correlação positiva e significativa entre fluxo expiratório e distância percorrida (r = 0,635 e p < 0,001). Conclusão: Os indivíduos obesos tiveram menor PFE e menor distância percorrida no TC6´, constatada correlação positiva entre essas duas variáveis. Diante de tal desfecho, a inclusão dos métodos avaliativos utilizados podem contribuir para melhor análise da condição respiratória e funcional dessa população e auxiliar na prescrição de exercícios.

A non-destructive measuring technique was applied to test major vine geometric traits on measurements collected by a contactless sensor. Three-dimensional optical sensors have evolved over the past decade, and these advancements may be useful in improving phenomics technologies for other crops, such as woody perennials. Red, green and blue-depth (RGB-D) cameras, namely Microsoft Kinect, have a significant influence on recent computer vision and robotics research. In this experiment an adaptable mobile platform was used for the acquisition of depth images for the non-destructive assessment of branch volume (pruning weight) and related to grape yield in vineyard crops. Vineyard yield prediction provides useful insights about the anticipated yield to the winegrower, guiding strategic decisions to accomplish optimal quantity and efficiency, and supporting the winegrower with decision-making. A Kinect v2 system on-board to an on-ground electric vehicle was capable of producing precise 3D point clouds of vine rows under six different management cropping systems. The generated models demonstrated strong consistency between 3D images and vine structures from the actual physical parameters when average values were calculated. Correlations of Kinect branch volume with pruning weight (dry biomass) resulted in high coefficients of determination (R2 = 0.80). In the study of vineyard yield correlations, the measured volume was found to have a good power law relationship (R2 = 0.87). However due to low capability of most depth cameras to properly build 3-D shapes of small details the results for each treatment when calculated separately were not consistent. Nonetheless, Kinect v2 has a tremendous potential as a 3D sensor in agricultural applications for proximal sensing operations, benefiting from its high frame rate, low price in comparison with other depth cameras, and high robustness.

Whole-genome sequencing projects are increasingly populating the tree of life and characterizing biodiversity1-4. Sparse taxon sampling has previously been proposed to confound phylogenetic inference5, and captures only a fraction of the genomic diversity. Here we report a substantial step towards the dense representation of avian phylogenetic and molecular diversity, by analysing 363 genomes from 92.4% of bird families-including 267 newly sequenced genomes produced for phase II of the Bird 10,000 Genomes (B10K) Project. We use this comparative genome dataset in combination with a pipeline that leverages a reference-free whole-genome alignment to identify orthologous regions in greater numbers than has previously been possible and to recognize genomic novelties in particular bird lineages. The densely sampled alignment provides a single-base-pair map of selection, has more than doubled the fraction of bases that are confidently predicted to be under conservation and reveals extensive patterns of weak selection in predominantly non-coding DNA. Our results demonstrate that increasing the diversity of genomes used in comparative studies can reveal more shared and lineage-specific variation, and improve the investigation of genomic characteristics. We anticipate that this genomic resource will offer new perspectives on evolutionary processes in cross-species comparative analyses and assist in efforts to conserve species.

The evaluation of thiamine and its derivative phosphate esters levels in pregnant women in rural communities can contribute not only for understanding the specific characteristics of this population regarding nutritional aspects, but also for clarifying the relations of psychiatric manifestations and a vitamin deficit. In the present work we assessed sociodemographic variables, psychiatric parameters and thiamine and its derivative in the whole blood of women in a rural, low-income community in Brazil. A case-control study was done. 94 women were divided in groups using the trimesters of pregnancy as a criterion: each trimester, 1st, 2nd and 3rd had 17, 37 and 38 women, respectively. A control group of non-pregnant women (n-39) was also included. Symptoms of anxiety and depression were assessed using the HAMA Scale and Beck Inventory, respectively. The thiamine and its phosphorylated derivatives concentrations were determined in whole blood samples using the HPLC method. The results suggest that physiological mechanisms linked to the metabolic pathways of thiamine may play a role in some neurobiological substrate involved in the regulation of emotional state. Thus, social vulnerability is identified as an important factor to be considered in the evaluation of the mental health of pregnant women living in rural communities.

Crop 3D modeling allows site-specific management at different crop stages. In recent years, light detection and ranging (LiDAR) sensors have been widely used for gathering information about plant architecture to extract biophysical parameters for decision-making programs. The study reconstructed vineyard crops using light detection and ranging (LiDAR) technology. Its accuracy and performance were assessed for vineyard crop characterization using distance measurements, aiming to obtain a 3D reconstruction. A LiDAR sensor was installed on-board a mobile platform equipped with an RTK-GNSS receiver for crop 2D scanning. The LiDAR system consisted of a 2D time-of-flight sensor, a gimbal connecting the device to the structure, and an RTK-GPS to record the sensor data position. The LiDAR sensor was facing downwards installed on-board an electric platform. It scans in planes perpendicular to the travel direction. Measurements of distance between the LiDAR and the vineyards had a high spatial resolution, providing high-density 3D point clouds. The 3D point cloud was obtained containing all the points where the laser beam impacted. The fusion of LiDAR impacts and the positions of each associated to the RTK-GPS allowed the creation of the 3D structure. Although point clouds were already filtered, discarding points out of the study area, the branch volume cannot be directly calculated, since it turns into a 3D solid cluster that encloses a volume. To obtain the 3D object surface, and therefore to be able to calculate the volume enclosed by this surface, a suitable alpha shape was generated as an outline that envelops the outer points of the point cloud. The 3D scenes were obtained during the winter season when only branches were present and defoliated. The models were used to extract information related to height and branch volume. These models might be used for automatic pruning or relating this parameter to evaluate the future yield at each location. The 3D map was correlated with ground truth, which was manually determined, pruning the remaining weight. The number of scans by LiDAR influenced the relationship with the actual biomass measurements and had a significant effect on the treatments. A positive linear fit was obtained for the comparison between actual dry biomass and LiDAR volume. The influence of individual treatments was of low significance. The results showed strong correlations with actual values of biomass and volume with R2 = 0.75, and when comparing LiDAR scans with weight, the R2 rose up to 0.85. The obtained values show that this LiDAR technique is also valid for branch reconstruction with great advantages over other types of non-contact ranging sensors, regarding a high sampling resolution and high sampling rates. Even narrow branches were properly detected, which demonstrates the accuracy of the system working on difficult scenarios such as defoliated crops.

The Pseudomonas genus, which includes environmental and pathogenic species, is known to present antibiotic resistances, and can receive resistance genes from multi-resistant enteric bacteria released into the environment via faecal rejects. This study was aimed to investigate the resistome of Pseudomonas populations that have been in contact with these faecal bacteria. Thus, faecal discharges originating from human or cattle were sampled (from 12 points and two sampling campaigns) and 41 Pseudomonas species identified (316 isolates studied). The resistance phenotype to 25 antibiotics was determined in all isolates, and we propose a specific antibiotic resistance pattern for 14 species (from 2 to 9 resistances). None showed resistance to aminoglycosides, tetracycline, or polymyxins. Four species carried a very low number of resistances, with none to ß-lactams. Interestingly, we observed the absence of the transcriptional activator soxR gene in these four species. No plasmid transfer was highlighted by conjugation assays, and a few class 1 but no class 2 integrons were detected in strains that may have received resistance genes from Enterobacteria. These results imply that the contribution of the Pseudomonas genus to the resistome of an ecosystem first depends on the structure of the Pseudomonas populations, as they may have very different resistance profiles.

BACKGROUND: The giant squid (Architeuthis dux; Steenstrup, 1857) is an enigmatic giant mollusc with a circumglobal distribution in the deep ocean, except in the high Arctic and Antarctic waters. The elusiveness of the species makes it difficult to study. Thus, having a genome assembled for this deep-sea-dwelling species will allow several pending evolutionary questions to be unlocked. FINDINGS: We present a draft genome assembly that includes 200 Gb of Illumina reads, 4 Gb of Moleculo synthetic long reads, and 108 Gb of Chicago libraries, with a final size matching the estimated genome size of 2.7 Gb, and a scaffold N50 of 4.8 Mb. We also present an alternative assembly including 27 Gb raw reads generated using the Pacific Biosciences platform. In addition, we sequenced the proteome of the same individual and RNA from 3 different tissue types from 3 other species of squid (Onychoteuthis banksii, Dosidicus gigas, and Sthenoteuthis oualaniensis) to assist genome annotation. We annotated 33,406 protein-coding genes supported by evidence, and the genome completeness estimated by BUSCO reached 92%. Repetitive regions cover 49.17% of the genome. CONCLUSIONS: This annotated draft genome of A. dux provides a critical resource to investigate the unique traits of this species, including its gigantism and key adaptations to deep-sea environments.

Plant modeling can provide a more detailed overview regarding the basis of plant development throughout the life cycle. Three-dimensional processing algorithms are rapidly expanding in plant phenotyping programmes and in decision-making for agronomic management. Several methods have already been tested, but for practical implementations the trade-off between equipment cost, computational resources needed and the fidelity and accuracy in the reconstruction of the end-details needs to be assessed and quantified. This study examined the suitability of two low-cost systems for plant reconstruction. A low-cost Structure from Motion (SfM) technique was used to create 3D models for plant crop reconstruction. In the second method, an acquisition and reconstruction algorithm using an RGB-Depth Kinect v2 sensor was tested following a similar image acquisition procedure. The information was processed to create a dense point cloud, which allowed the creation of a 3D-polygon mesh representing every scanned plant. The selected crop plants corresponded to three different crops (maize, sugar beet and sunflower) that have structural and biological differences. The parameters measured from the model were validated with ground truth data of plant height, leaf area index and plant dry biomass using regression methods. The results showed strong consistency with good correlations between the calculated values in the models and the ground truth information. Although, the values obtained were always accurately estimated, differences between the methods and among the crops were found. The SfM method showed a slightly better result with regard to the reconstruction the end-details and the accuracy of the height estimation. Although the use of the processing algorithm is relatively fast, the use of RGB-D information is faster during the creation of the 3D models. Thus, both methods demonstrated robust results and provided great potential for use in both for indoor and outdoor scenarios. Consequently, these low-cost systems for 3D modeling are suitable for several situations where there is a need for model generation and also provide a favourable time-cost relationship.

Arid environments provide ideal ground for investigating the mechanisms of adaptive evolution. High temperatures and low water availability are relentless stressors for many endotherms, including birds; yet birds persist in deserts. While physiological adaptation probably involves metabolic phenotypes, the underlying mechanisms (plasticity, genetics) are largely uncharacterized. To explore this, we took an intraspecific approach that focused on a species that is resident over a mesic to arid gradient, the Karoo scrub-robin (Cercotrichas coryphaeus). Specifically, we integrated environmental (climatic and primary productivity), physiological (metabolic rates: a measure of energy expenditure), genotypic (genetic variation underlying the machinery of energy production) and microbiome (involved in processing food from where energy is retrieved) data, to infer the mechanism of physiological adaptation. We that found the variation in energetic physiology phenotypes and gut microbiome composition are associated with environmental features as well as with variation in genes underlying energy metabolic pathways. Specifically, we identified a small list of candidate adaptive genes, some of them with known ties to relevant physiology phenotypes. Together our results suggest that selective pressures on energetic physiology mediated by genes related to energy homeostasis and possibly microbiota composition may facilitate adaptation to local conditions and provide an explanation to the high avian intraspecific divergence observed in harsh environments.

Bivalve molluscs of the genus Mytilus are considered a model organism in ecotoxicology and are known to be well adapted to marine ecosystems affected by multiple anthropogenic factors, including pollution. In order to assess whether pollution interferes with the reproductive success of Mytilus and affects the diversity within and between populations, we sequenced the transcriptomes of 72 individuals from 9 populations of Mytilus galloprovincialis collected along a ca. 130-km north-south transect on the Western coast of the Iberian Peninsula. We found that polluted areas are acting as a barrier to gene flow, potentially because of the detrimental effect of anthropogenic chemicals on larvae carried from more pristine environments. Furthermore, we observed an increase in genetic diversity in populations from polluted site, which could be indicative of higher mutagenicity driven by the environment. We propose that a microevolutionary perspective is essential for a full characterization of human activities on the dispersal of M. galloprovincialis and that it should be incorporated into management, and conservation plans and policies in the context of the effects of pollution on populations.

Thiamine deficiency (TD) has been used as an experimental model in rodents to study the molecular mechanisms of neurodegeneration and its association with behavioral changes. The aims of the present study were to investigate the spatial cognitive performance of pyrithiamine-induced thiamine deficiency (PTD) in adult male rats and disclose the thalamic proteome alterations caused by a severe TD episode. After the onset of the neurological signs, such as seizure and/or loss of righting reflex, the TD treatment was interrupted. Following 15 days of recovery, all rats were submitted to the spatial cognitive tasks in the Morris Water Maze (MWM). The results show that the PTD rats exhibited deficits during the learning process, which was reverted by repeated training. However, despite the spatial cognitive recovery, some protein changes were not reversible. The proteomic analysis, using label-free quantification, revealed deregulation of 183 thalamic proteins. Using bioinformatic tools, these proteins were categorized according to Gene Ontology functional annotation and metabolic pathways. We show that a severe TD affects proteins involved in different biological processes, such as, oxidative stress, neurotransmitter synthesis and synaptic vesicle cycle. These could explain the outcome in neurotransmitter release changes caused by TD, previously observed by our group and by other authors. These findings disclose the role of key proteins and metabolic pathways probably involved in the neurodegeneration process induced by TD. These proteins represent relevant molecular targets for future studies focusing also on the molecular basis of selective vulnerability of some brain areas to TD insult.

Sensing advances in plant phenotyping are of vital importance in basic and applied plant research. Plant phenotyping enables the modeling of complex shapes, which is useful, for example, in decision-making for agronomic management. In this sense, 3D processing algorithms for plant modeling is expanding rapidly with the emergence of new sensors and techniques designed to morphologically characterize. However, there are still some technical aspects to be improved, such as an accurate reconstruction of end-details. This study adapted low-cost techniques, Structure from Motion (SfM) and MultiView Stereo (MVS), to create 3D models for reconstructing plants of three weed species with contrasting shape and plant structures. Plant reconstruction was developed by applying SfM algorithms to an input set of digital images acquired sequentially following a track that was concentric and equidistant with respect to the plant axis and using three different angles, from a perpendicular to top view, which guaranteed the necessary overlap between images to obtain high precision 3D models. With this information, a dense point cloud was created using MVS, from which a 3D polygon mesh representing every plants' shape and geometry was generated. These 3D models were validated with ground truth values (e.g., plant height, leaf area (LA) and plant dry biomass) using regression methods. The results showed, in general, a good consistency in the correlation equations between the estimated values in the models and the actual values measured in the weed plants. Indeed, 3D modeling using SfM algorithms proved to be a valuable methodology for weed phenotyping, since it accurately estimated the actual values of plant height and LA. Additionally, image processing using the SfM method was relatively fast. Consequently, our results indicate the potential of this budget system for plant reconstruction at high detail, which may be usable in several scenarios, including outdoor conditions. Future research should address other issues, such as the time-cost relationship and the need for detail in the different approaches.

RESUMO

Depression is a complex condition, and its classical biological/psychosocial distinction is fading. Current guidelines are increasingly advocating psychotherapy as a treatment option. Psychodynamic psychotherapy models encompass a heterogeneous group of interventions derived from early psychoanalytic conceptualizations. Growing literature is raising awareness in the scientific community about the importance of these treatment options, as well as their favorable impact on post-treatment outcomes and relapse prevention. Considering the shifting paradigm regarding treatment of depressive disorder, the authors aim to provide a brief overview of the definition and theoretical basis of psychodynamic psychotherapy, as well as evaluate current evidence for its effectiveness.
